Rabies is considered a fatal disease once clinical symptoms have developed. The aim of this study was to evaluate epidemiological aspects and immune response in patients attacked by domestic and wild animals and subjected to post-exposure rabies treatment with equine serum and associated vaccine. Thirty-three patients were evaluated; they were between 13 and 65 years old, 75.8% were male and 24.2% female, and from the Botucatu neighborhood. Twenty healthy control individuals with the same age range were also studied. Specific antibodies to equine immunoglobulins and IFN-gamma, IL-2, IL-4, and IL-10 production were evaluated by ELISA. IgM, IgE, IgG and subclasses, and rabies virus antibodies serum levels were determined by nephelometry and... |

Bats are very interesting animals: they are the unique flying mammals, have developed a highly sophisticated echolocation system, and have become specialized to eat different types of diets. Hematophagous (vampire) bats are those specialized to feed solely on blood and have served as a source of inspiration for researchers as well as for writers. Vampire bat attacks on humans have moved from the realm of science fiction to reality in Latin America and bats (including non-hematophagous ones) have assumed an important role in the transmission of rabies virus to humans. This article discusses the emerging role of bats as rabies virus transmitters, with particular emphasis on the role of hematophagous bats in the epidemiology of human rabies in Latin America.... |

Abstract Background: Between 40,000-70,000 people die yearly of rabies, an incurable disease. Besides post-bite vaccination, no treatment is available for it. Methods: First, virus dilution for antiviral effects in mice was determined. Then, animals were treated as follows: control (NaCl 250 µL/animal/day); bufotenine (0.63, 1.05 and 2.1 mg in 250 µL of NaCl/animal/day); rabies (10-6,82CVS dilution); and test (10-6,82 CVS dilution and bufotenine, in the above-mentioned doses). Animals were observed daily for 21 days or until the 3rd stage of rabies infection. Twitch-tension and liposome studies were applied to understand the possible interaction of bufotenine with receptors, particularly acetylcholine. Results: Bufotenine was able to increase the... |

Background Rabies is a fatal zoonotic neglected disease that occurs in more than 150 countries, and kills more than 55.000 people every year. It is caused by an enveloped single stranded RNA virus that affects the central nervous system, through an infection initiated by the muscular nicotinic acetylcholine receptor, according to many authors. Alkaloids, such as acetylcholine, are widespread molecules in nature. They are present in numerous biological fluids, including the skin secretion of many amphibians, in which they act (together with proteins, peptides and steroids) as protection agents against predators and/or microorganisms. Among those amphibians that are rich in alkaloids, there is the genus Rhinella. Methods Bufotenine was isolated from Rhinela... |

ABSTRACTWe have evaluated the efficacy of short-interfering RNAs targeting the nucleoprotein gene and also the brain immune response in treated and non-treated infected mice. Mice were inoculated with wild-type virus, classified as dog (hv2) or vampire bat (hv3) variants and both groups were treated or leaved as controls. No difference was observed in the lethality rate between treated and non-treated groups, although clinical evaluation of hv2 infected mice showed differences in the severity of clinical disease (p = 0.0006). Evaluation of brain immune response 5 days post-inoculation in treated hv2 group showed no difference among the analyzed genes, whereas after 10 days post-inoculation there was increased expression of 2',5'-oligoadenylate synthetase... |

ABSTRACTRabies virus (RABV) isolated from different mammals seems to have unique characteristics that influence the outcome of infection. RABV circulates in nature and is maintained by reservoirs that are responsible for the persistence of the disease for almost 4000 years. Considering the different pattern of pathogenicity of RABV strains in naturally and experimentally infected animals, the aim of this study was to analyze the characteristics of RABV variants isolated from the main Brazilian reservoirs, being related to a dog (variant 2),Desmodus rotundus (variant 3), crab eating fox, marmoset, and Myotis spp. Viral replication in brain tissue of experimentally infected mouse was evaluated by two laboratory techniques and the results were compared to... |

The National Programme for Rabies Prevention aims to maintainimmunogenic protective levels in vaccinated animals, with titersof neutralizing antibodies (VNA) ≥ 0.5 IU / mL. The aim of thisstudy was to evaluate, according to age, race, and the periodof vaccination and blood collection, the immune response indogs that received a single dose of rabies vaccine virus in cellculture. In this study 432 samples received at the Pasteur Instituteof São Paulo in the triennium 2009-2011 were analyzed. Datawere analyzed and the evaluation of the VNA to rabies virus wasperformed by rapid fluorescent inhibition test foci (RFFIT). Of thetotal samples analyzed, 21.76% did not have protective titers.Among these, 67.02% samples were considered puppies andwhen the interval... |

Rabies is a zoonotic disease that affects all mammals and leads to more than 55,000 human deaths every year, caused by rabies virus (RABV) (Mononegavirales: Rhabdoviridae: Lyssavirus). Currently, human rabies treatment is based on the Milwaukee Protocol which consists on the induction of coma and massive antiviral therapy. The aim of this study was to assess the decrease in the titer of rabies virus both in vitro and in vivo using short-interfering RNAs. To this end, three siRNAs were used with antisense strands complementary to rabies virus nucleoprotein (N) mRNA. BHK-21 cells monolayers were infected with 1000 to 0.1 TCID50 of PV and after 2 hours the cells were transfected with each of tree RNAs in separate using Lipofectamine-2000. All three siRNAs... |

In this study, we compared the levels of neutralizing antibodies induced by inactivated rabies vaccine in cattle by using three alternative immunization procedures. Forty-five bovines (breed nelore) were then organized in three groups (A, B and C, with 15 animals/group). Group A received only one vaccine dose at day zero and Group B received the first dose at day zero and then another dose at day 30 (early booster). Group C was also immunized with two doses; however, the booster was postponed until day 180 after the first dose (delayed booster). Blood samples were withdrawn at days zero (before the first dose) and 30, 210, 390, and 540 after the beginning of immunization and the antibody titers were evaluated by mouse neutralization test. The protocol used... |

Background Rabies, a zoonosis found throughout the globe, is caused by a virus of theLyssavirus genus. The disease is transmitted to humans through the inoculation of the virus present in the saliva of infected mammals. Since its prognosis is usually fatal for humans, nationwide public campaigns to vaccinate dogs and cats against rabies aim to break the epidemiological link between the virus and its reservoirs in Brazil.Findings During 12 months we evaluated the active immunity of dogs first vaccinated (booster shot at 30 days after first vaccination) against rabies using the Fuenzalida-Palácios modified vaccine in the urban area of Botucatu city, São Pauto state, Brazil. Of the analyzed dogs, 54.7% maintained protective titers (≥0.5 IU/mL) for 360 days... |

The relationship among the phenotypes resistance to infection, virus replication in the brain and isotype production was investigated in genetically modified High (H) or Low (L) antibody responder mouse lines. Although they express the same innate susceptibility to rabies infection, these lines differ as to different viral replication rates in the central nervous system and L mice showed a higher permissible state. After intramuscular infection with the Pasteur rabies strain (PV), the H-L interline differences on the earlier stage of virus replication were 1000 and 80 folds on days 5 and 6, respectively. The isotype profile in sera of the experimentally infected mice reflected an interline difference of 25 folds for IgG2a throughout the infection period,... |

Rabies is a viral zoonotic infectious disease that affects mammals and is caused by genotypes/species of the Lyssavirus genus (Rhabdoviridae, Mononegavirales), with the genotype 1 (classic rabies virus - RABV) being the most prevalent. Despite continuous efforts, rabies is still an incurable disease that causes thousands of deaths amongst humans worldwide. Due to a wide range of hosts and the different evolutionary paths of RABV in each host, several host-specific variants have arisen in an ongoing process. The result of RABV replication in nervous tissues may lead to two opposite clinical outcomes, i.e., paralytic/dumb form and encephalitic/furious one. The paralytic form creates dead-end hosts mainly amongst herbivores, while the furious form of the... |

Hamsters orally inoculated with ERA and PV strains of rabies virus were sacrificed at 24, 48, 72 hours, 21 and 30 days after inoculation. Brain fragments were examined by Fluorescent Antibody test (FAT) and heminested PCR (hn-PCR). Fragments from stomach, blood, heart, and lung were examined only by hn-PCR. Sera of other hamsters, similarly inoculated, obtained at 30th day after inoculation were submitted to mouse neutralization test. The hamsters were challenged intracerebrally with CVS strain with 10(2.7)mouse LD50/0.03mL, 45 days after inoculation. Brains examined by FAT were negative. The hn-PCR detected the presence of rabies virus RNA in the lung of one animal inoculated with ERA, and in the brain, stomach, blood, and lung of PV-infected animals. The... |

This report describes pneumomediastinum and subcutaneous emphysema in a bull with paralytic rabies. The bull displayed severe swelling in the head, neck, chest and forelimbs extending to the ventrum, including the prepuce, 7 days after the development of neurological signs. At necropsy, extensive subcutaneous emphysema was observed in the neck region, abdominal wall and proximal forelimbs. The lungs failed to collapse and had severe emphysema with bullae. Rabies was suspected due to an ongoing outbreak at the farm and was confirmed by histologic lesions and immunohistochemistry. Similar to humans, pneumomediastinum and subcutaneous emphysema should be considered as an unusual complication of rabies in cattle. |

Official reporting of rabies in Nigeria started in 1912. The National Veterinary Research Institute (NVRI), Vom, within the last 77 years, confirmed 4809 cases of animal rabies in Nigeria. Rabies control through immunization programs has crashed woefully and consequently, the rabies situation has become chaotic and confounding. Locally, only 2,137,615 doses of dog anti-rabies vaccines were produced between 1956 and 2005 (average 43,625 per year) by NVRI, Vom. Dog population in Nigeria is currently estimated at 8 million. In the last 20 years of research and follow ups on rabies and associated problems, only 10% of the dogs’ population received anti-rabies immunization. Within the immunized dogs, rabies outbreaks occurred frequently. Evidences on the trend... |
